▎  TNF-α 概述


肿瘤坏死因子α(TNF-α),他的基因编码一种多功能促炎细胞因子,该因子属于肿瘤坏死因子(TNF)超家族。这种细胞因子主要由巨噬细胞分泌。它可以通过其受体TNFRSF1A/TNFR1和TNFRSF1B/TNFBR进行结合,并发挥功能。这种细胞因子参与多种生物过程的调节,包括细胞增殖、分化、凋亡、脂质代谢和凝血。这种细胞因子与多种疾病有关,包括自身免疫性疾病、胰岛素抵抗、银屑病、类风湿性关节炎、强直性脊柱炎、结核病、常染色体显性多囊肾病和癌症。该基因的突变会影响脑型疟疾、感染性休克和阿尔茨海默病的易感性。小鼠中的基因敲除研究也表明这种细胞因子具有神经保护作用。

▎  用途


该试剂盒采用“一步”夹心法酶联免疫吸附试验(ELISA)体外定量检测血清、血浆、组织、细胞或其他相关生物液体中TNF-α浓度。


  检测原理


将目标抗体包被于96孔微孔板中,制成固相载体,向微孔中分别加入标准品或标本,其中的目标连接于固相载体上的抗体结合,然后加入辣根过氧化物酶标记的抗体,将未结合的抗体洗净后再次彻底洗涤后加入TMB底物显色。TMB在过氧化物酶的催化下转化成蓝色,并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的目标呈正相关。用酶标仪在450nm波长下测定吸光度(O.D.值),计算样品浓度。

选取3个不同批次的试剂盒分别对低、中、高值定值样本定量测定,每个样本使用同一试剂盒重复测定8次,分别计算不同浓度样本的平均值及SD值


▎  精密度


精密度用样品测定值的变异系数CV表示。CV(%)=SD/mean×100  CV<10% Inter-批间差: CV<13%


▎  稳定性


经测定,试剂盒在有效期内按推荐温度保存,其活性降低率小于5%。
为减小外部因素对试剂盒破坏前后检测值的影响,实验室的环境条件需尽量保持一致,尤其是实验室内温度、湿度及温育条件。其次由同一实验员来进行操作可减少人为误差。

  实验所需自备物品

1、450±10nm滤光片酶标仪(建议仪器使用前预热)  

2、高精度加样器及枪头:0.5-10uL、2-20uL、20-200uL、200-1000uL.   

3、Eppendorf 移液器.   

4、蒸馏水或去离子水.   

5、脱脂棉吸水纸.   

6、37℃恒温箱.
